Effect of Acetosyringone and Callus Age on Transformation for Scutellum –derived Callus of Rice

چکیده
An efficient and reproducible protocol for Agrobacteriummediated transformation of an agronomically useful Oryza sativa variety, Pusa Basmati1 (IET10364) has been standardized. Although several reporters have been claimed the Agrobacteriummediated transformation but here we studied the effect of acetosyringone and age of callus on transformation frequency for scutellumderived callus of Pusa basmati1. By optimizing these two parameters for Agrobateriummediated transformation, we can produce an efficient and reproducible protocol. The callus derived from mature rice seeds were used as explants for transformation studies. Callus were co-cultured with Agrobacterium tumefaciens EHA 105 harboring a binary vector p35SGUSINT that carried Betaglucuronidase (uid A) and It contains gene npt II, determining resistance to kanamycin, under the control of the constitutive (nos) promoter. Maximum Agrobacteriummediated transformation frequency was observed at 350μM/l acetosyringone with 35 days old callus.

Studies on the Effect of Genotype and Explant Type on Callogenesis and Organogenesis in Indica Rice
The purpose of the research study is to select the best rice explant and variety for Agrobacterium mediated genetic transformation studies. In vitro callus induction and regeneration frequency of two explants i.e. immature embryo and mature grains of 9 rice varieties (Oryza sativa L.) was investigated by culturing explants source on N6 media supplemented with 2, 4-D at 2mg/l for callus inductio...
متن کاملTransient expression of -glucuronidase gene in indica and japonica rice (Oryza sativa L.) callus cultures after different stages of co-bombardment
Rapidly growing embryogenic calli of different ages derived from the scutellum of mature seed embryos of indica rice (Rasi) and mature seeds of japonica rice (Taipei 309) was biolistically transformed. Plasmid pUCGUS containing the uidA gene encoding -glucuronidase was used to optimize transformation conditions using various combinations of helium pressure, target distance and gap distance. Pla...
متن کاملProduction of herbicide-tolerant zoysiagrass by Agrobacterium-mediated transformation.
Herbicide-resistant zoysiagrass (Zoysia japonica Steud.) has been developed by Agrobacterium-mediated transformation. A callus-type transformation system was established by optimizing several factors that affect the rate of transformation, including co-cultivation period and concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D), CaCl2 and acetosyringone. Maximal GUS expression was observed w...
متن کاملTransgenic plant production using particle bombardment in Egyptian barley
A system for Egyptian barley (Hordeum vulgare L. cv Giza 123) transformation via biolistic bombardment has been established. Immature embryos 1.0-1.5 mm in length were aseptically isolated from barley plants cultivated in the open field. Embryos were then cultured on callus induction medium with scutellum side up. Scutellum derived calli were bombarded with the plasmid pAHC25, containing the ma...
